Is rain water safe to drink straight from a barrel?

Rain water is not safe to drink straight from a barrel. The rain itself starts out close to distilled, but by the time it has crossed a roof, a gutter and a storage tank it has picked up bird droppings, roof debris, dust and whatever the roofing material sheds. What treatment would actually be needed

Making collected rain water genuinely drinkable takes a sequence, not a single filter. A first flush diverter dumps the dirtiest opening minutes of a storm. Sediment filtration removes grit and organic debris. Carbon deals with taste, odor and organic compounds shed by the roof. Then disinfection, ultraviolet or chemical, addresses the microbiological risk, and ultraviolet only works on water already clear enough for the light to pass through. Rain is also very soft and low in dissolved minerals, which can make it aggressive toward copper plumbing, so pH may need correcting too.

Uses that do not need any of that

Collected rain water is genuinely useful for things that never reach a mouth: garden and landscape irrigation, washing a car, topping up outdoor cleaning tasks. It is naturally soft, so it leaves fewer spots than a hard municipal supply and suits plants that dislike hard water. Keep the barrel covered and screened to keep mosquitoes and debris out, and keep the catchment plumbing entirely separate from your drinking supply.
